| PRESENTATION | After extraction from infected cells, the virus is concentrated by ultracentrifugation and then partially purified by centrifugation on a sucrose density gradient. The antigen is resuspended and homogenised. The resulting antigen preparation consists of a high concentration of virus and viral components as well as some cellular material suspended in 100 mM glycine buffer, pH 9.5 |
| PROTEIN CONCENTRATION | mg/ml by Bradford (Biorad) assay using BSA as standard. |
| INACTIVATION | By treatment with β-propiolactone. Infectivity confirmed negative by absence of growth under original cell culture conditions. |
